Her father, Anil Ganguly, was a film director, and her mother, Rajni Ganguly, is a film producer. She has a brother named Vijay Ganguly, who is an actor and producer. Rupali married Ashwin K Verma on 6 February 2013, and they have a son named Rudransh Verma.
She completed her schooling at Kolkata and studied hotel management degree. After completing her graduation, she made her debut in the television serial Sukanya. She also participated as a contestant in the popular reality shows Bigg Boss in 2006. Ganguly is currently playing the lead role in Hindi Serial Anupamaa aired on Star Plus.
